Resource competition shapes CRISPR-mediated gene activation (opens in new tab)
Aravind et al. show that competition for resources limits CRISPRa efficiency producing a biphasic on-target response. These effects are only moderately alleviated by increasing resource levels to the maximum limit tolerated by the cell. A reaction-network model predicts these effects and is a tool for optimizing multi-module CRISPRa networks.
