Lee et al. present a screen of inhibitory CAR designs and identify LIR1 as the most promising intracellular signaling domain. The LIR1 iCAR functions in both T and NK cells, operates in cis, and transduces signaling via SHP1. iCAR can also reduce exhaustion phenotypes and potentially enhance antitumor responses.
